Will these work on the roof mounting threads?

Are the threads long enough and the right size for a 2020 250?

maybe, yes. I'm not sure they'd cover the access hole to the threads though...
Thing is... to seal the hole, you need something to cover it. Depending on the thickness of whatever you put between those and the hole will determine if the thread is long enough. I'd lean toward "not ideal"...
People have used ones that look like those in past forum threads.
look on www.mcmaster.com for way more options, sealing washers, etc. but yes you can do that.
sealing washers
If you use an eye nut and some threaded rod (or cut the thread off a bolt) then will be possible to control the orientation of the eye and mount of compression of the washer... with some trial and error "fiddling about".
Threaded rod rusts badly but I guess you can paint it.

Yup for the basic hardware store stuff, but It comes in stainless .
Also in this application to maintain orientation of bolt, should use CA glue or red threadlocker on that interface , and the rest is inside/ beneath the sealing washer where I'd use a blue thread locker.

You can buy stainless steel all-thread. Not really that hard to come by.
Or you could just buy the eye bolt and be done with it, Nice and simple.
People have been putting eye bolts on the roof of the transit for years.
