The driver side door lock switch when pressed does not do anything however if I press the key fob, remote keyless entry, or passenger door lock switch all the locks open. I changed the switch in the drivers door and it still does not work. Is there a fuse, relay or programing issue I am missing. It just randomly quit working one day. Any ideas are appreciated.

This might help:
Connector C505 is in the drivers door, it connects the door lock switch to the wiring harness in the door.
Connector C339 is in the drivers door jamb, it's mounted to the A piller with a rubber bellows.

I noticed that if I'm in the van and lock the doors with the remote, neither door unlock switch will unlock the doors. Kind of a safety issue in my mind, so I make sure when I park I put the keys away and lock the doors with the door switch so if I need to unlock them in the middle of the night I know the door switch will work.

Not sure if that is your issue or it never operates, in which case my problem isn't yours.
